Tumors have higher glucose consumption compared with normal tissues due to anaerobic glycolysis for energy production. Therefore, accurate detection of glucose distribution in vivo may be a promising method for tumor tracking. In this study, an initiator and a dye are encapsulated into an amphiphilic polymer carrier as chemiluminescence nanoparticles (CLNPs) by the nanoprecipitation method, and glucose oxidase (GOx) is loaded on the surface of the nanoparticles to design a chemiluminescence probe responding to glucose. The GOx-modified CL nanoparticles (CLNPs-GOx) can quantitatively characterize glucose concentration with a low detection limit and good specificity, which enables not only recognizing the tumors from normal tissues but also identifying the development stage of cancer cells with an in vivo imaging approach. By consuming glucose and generating cytotoxic H2O2, the prepared probe also had an inhibitory effect on tumor growth. These results indicate that CL probes have better sensitivity and selectivity for glucose detection compared to traditional fluorescence imaging methods, and CLNPs-GOx have potential applications for cancer diagnosis and treatment.
