• We discover that SlanMo have the capacity to migrate along a gradient of ICs. • This IC-induced, gradient-dependent directed migration is not shared by other leukocyte subtypes. • FcγRIII (CD16) is of primary importance for high-affinity binding of IC and the induction of directed migration. • Phosphoproteomic analysis of migrating SlanMo, identified ROCK, Lyn, crk, FAK, and moesin as relevant signaling proteins. • Transcriptomic studies on migrating SlanMo reveal a lupus-associated activation signature.
