• A hybrid deconvolution method for 3D fluorescence microscopy is proposed. • Effectively subtracts out-of-focus background prior to RL refinement. • The method minimizes defocus halos and improves structural fidelity. • Drives scattering background to near-zero in thick-tissue neuronal imaging.
