• First-principles mathematical model predicts tablet weight variability from particle size distribution (PSD) and tooling geometry. • Single-parameter calibration accurately captures weight variability across a wide design space spanning PSD, tooling dimensions and tablet weight. • Framework enables prediction and control of tablet weight variability using physical properties of blends and tooling dimensions, instead of empirical heuristics.
