• Force modulates ADAMTS-5–aggrecan binding through multiple stabilization switches. • Stable binding states identified at 0, 100, 400, and 900 pN by molecular simulations. • Distinct residues mediate force-dependent binding via hydrogen bonds and salt bridges. • A multi-switch mechanism explains non-linear loading effects on aggrecan degradation. • Provides a molecular basis for protective vs. degradative roles of cartilage loading.
