• A tumor-targeted, aptamer-based, photocleavable DNA nanoflower (TAPE) is constructed for spatiotemporally controlled imaging of intracellular ATP in living cells. • The system integrates AS1411 aptamer-based tumor targeting and cholesterol modification, achieving up to 98.5% uptake efficiency in cancer cells. • A photocleavable linker enables a dual-lock “UV + ATP” activation logic, effectively suppressing non-specific signals before light triggering. • TAPE allows selective monitoring of drug-induced ATP fluctuations in living tumor cells.
