• A regulatory T cell-biased IL-2 mutein, Fc.mut24, selectively expands and enhances Tregs, promoting a tolerogenic immune environment in the lung. • Prophylactic Fc.mut24 treatment prevents the development of allergen-specific Th2 cells and allergic airway inflammation; this protection requires Tregs and is maintained even after allergen rechallenge. • Fc.mut24 treatment of mice after allergen sensitization is unable to inhibit allergen-specific Th2 cells and attenuate allergic airway inflammation, demonstrating a crucial early window of Treg-mediated tolerance induction.
