In this work, an alternative method for estimating hemoglobin from the thermal diffusivity values of erythrocyte supernatant is validated. Thermal values of test samples were studied and correlated with known hemoglobin concentrations. From the in vitro study, blood hemolysates were obtained after incubating erythrocytes with different concentrations of nanoparticles; in this case, SiO2-Au was used as a model. Thermal diffusivity values in hemolysates ranged from 10.20 +/- 0.28 x 10(-4) cm(2)/s to 11.98 +/- 0.14 x 10(-4) cm(2)/s. These values, together with a calibration curve, allowed hemoglobin quantification at low concentrations, up to 0.050 and 0.090 g/dl. According to UV-vis and thermal diffusivity results, hemoglobin release showed a linear behavior. This work suggests that the thermal diffusivity of hemolysates is closely related to hemoglobin concentration, making thermal lens spectroscopy an optimal technique for future applications in cytotoxic assays.
