• 40% ethanol was identified as a safe, effective, and low-cost solvent for quantitative extraction of betalain. • pGreenII 0800-RUBY vectors were constructed with a preserved multiple cloning site, enabling insertion of promoters. • pEAQ was validated as the optimal effector vector with the highest expression efficiency among four commonly used vectors.
