Adenosine 2A receptor drives microglial efferocytosis to accelerate white matter repair and functional recovery after stroke

Stroke produces cellular and myelin debris that is removed through a process called efferocytosis by microglia, the resident phagocytes of the brain. Deng et al. found that the adenosine 2A receptor (A2AR) in microglia was key for efficient efferocytosis. Microglial-specific deficiency in A2AR impaired remyelination after experimentally induced stroke in mice without affecting the extent of the initial injury. Conversely, micelles containing an A2AR agonist improved remyelination as well as cognitive and motor recovery after stroke but not if microglia were depleted or if microglia lacked A2AR. These results identify A2ARs in microglia as a potential pharmacological target to stimulate white matter repair and improve neurological outcomes after stroke. —Wei Wong
