Abstract

• This study introduces the "AIE-Pharmacology" paradigm by identifying the natural flavonoid Acacetin as a novel intrinsic AIEgen with ESIPT characteristics, enabling self-monitored therapy for asthenozoospermia. • The engineered EVs@N+A nanoplatform successfully overcomes the blood-testis barrier (BTB) to co-deliver Acacetin and NMN, leveraging the inherent biocompatibility of extracellular vesicles for targeted testicular accumulation. • This dual-therapy strategy synergistically rescues sperm motility and spermatogenesis by simultaneously inhibiting ferroptosis via the Nrf2/GPX4 pathway and rejuvenating cellular metabolism through NAD+-dependent ATP restoration.
