• Elevated L-2HG and reduced α-KG drive CD90+ FAP dysfunction in diabetic skeletal muscle. • The α-KG/L-2HG imbalance inhibits TET2, causing CD90+ FAP chromatin hypermethylation. • TET2-dependent epigenetic remodeling triggers aberrant asymmetric division related polarity program in CD90+ FAPs. • Dietary α-KG restores CD90+ FAP symmetry and alleviates muscle fibrosis in T2DM mice. • Targeting the α-KG-TET2 axis offers a metabolic therapy for diabetic muscle fibrosis.
