• ALPK1 is significantly upregulated in cardiac hypertrophy. • ALPK1 knockdown effectively suppresses Ang Ⅱ-induced NF-κB/NLRP3 inflammasome-mediated cardiomyocyte pyroptosis and hypertrophy. • ALPK1 over-expression promotes cardiomyocyte hypertrophy by activating NF-κB/NLRP3 inflammasome-mediated pyroptosis. • Targeting ALPK1 may provide new therapeutic strategies for cardiac hypertrophy.
