Amyloidogenic amylin signaling links diabetes to dementia pathogenesis

Amyloidogenic human amylin has emerged as a potential link between type-2 diabetes and Alzheimer’s disease. Pathological amylin signaling may connect peripheral metabolic stress with neurodegeneration. Chronic amylin receptor–cAMP–protein kinase A signaling disrupts cerebral energy metabolism and proteostasis. Experimental studies implicate amyloidogenic amylin in Alzheimer’s disease-related pathological pathways. Lowering circulating amyloidogenic amylin mitigates Alzheimer’s disease-like pathology in preclinical models. Selective modulation of pathogenic amylin signaling offers a new pharmacological strategy for Alzheimer’s disease.
