• Self-propelled PtNPs@COF accelerates PPIA binding via O2 generation. • DNAzyme release and CHA cascade amplify signal for ultrasensitive detection. • Detection limit of 0.330 pg/mL and wide linear range from 1 pg/mL to 10 μg/mL. • Reliably distinguishes lung adenocarcinoma patients from healthy controls.
