• α3β1-MEK/ERK regulates alternative polyadenylation of the Mmp9 gene during tissue remodeling. • α3β1-MEK/ERK induces cleavage stimulation factor CSTF3, a regulator of alternative polyadenylation. • DaPars2 analysis reveals that α3β1 regulates genome-wide alternative polyadenylation. • A novel α3β1-MEK/ERK-CSTF3 axis controls post-transcriptional gene regulation.
