An Off-On Peroxynitrite Fluorescent Probe Deigned by the Selective Oxidation of Nitrophenylglyoxylic Acid Ester

An off-on fluorescent probe (HCBO-NO2) for detecting peroxynitrite (ONOO−) was designed. The probe has a fluorophore combining an intramolecular charge transfer (ICT) system and a specific sensing site with the nitro to suppress fluorescence. By utilizing the oxidation properties of ONOO−, the dicarbonyl structure in nitrobenzoyl formate is oxidized and degraded, thereby eliminating the nitro part in the probe and achieving fluorescence recovery. The good ICT effect ensures that the fluorescent group has sufficient emission redshift to 590 nm. The detection limit of the probe HCBO-NO2 is as low as 26 nM, with high sensitivity and stability in the physiological range. Live-cell studies have shown that the probe can effectively detect both exogenous and endogenous ONOO, indicating that the probe HCBO-NO2 has potential value in practical biological applications.
