Anoikis resistance as a key driver of glioma malignancy: From ECM detachment to potential druggable targets

• Gliomas, especially glioblastoma, exhibit marked invasiveness, recurrence, and therapeutic resistance. • Detachment or dysregulated adhesion to the extracellular matrix (ECM) triggers anoikis, a specialized apoptotic process in mammalian cells. • Anoikis resistance is cooperatively mediated by dysregulated survival-related regulators, autocrine/paracrine signaling, protective autophagy, epithelial-mesenchymal transition (EMT), and metabolic reprogramming. • Anoikis resistance fuels glioma stemness, local infiltration, recurrence, and even metastatic dissemination. • Preclinical evidence indicates that pharmacological inhibition of specific mediators of anoikis resistance can restore anoikis sensitivity in glioma cells, presenting a promising therapeutic strategy.
