• Ionizing radiation for head and neck cancer damages surrounding structures. • Salivary gland damage lead to hyposalivation and chronic dry mouth. • Insertion of aquaporin-1 (AQP1) in the gland epithelium is a promising therapy. • Adenoviruses have successfully been used to deliver AQP1 to human salivary glands. • Adeno-associated viruses are a less immunogenic option for gene delivery.
