• The detection rate of AL-I in wheat samples was 100%, suggesting cumulative exposures could matter. • Much lower long-term toxicity of AL-1 than AA-1 in mice. • AL-I did not cause significant nephrotoxicity or obvious mutagenic effects, which was related to metabolism in body. • Mild renal interstitial collagen deposition may occur in AL-I-H group after a long period of discontinuation. • Remain vigilant about the potential nephrotoxicity risks of herbs containing AL-I and their environmental pollution risks.
