• XRF imaging showed that proteoglycan sulfation decreases during cartilage maturation • ARSI is expressed where cartilage sulfation decreases • Biochemical analyses show that ARSI is a novel endosulfatase of chondroitin-4-sulfate at acidic pH • ARSI localizes to lysosomes, which are disrupted in Arsi-/- chondrocytes • Maturation markers are upregulated in Arsi-/- chondrocytes
