• AFM mapping shows that surface remodeling peaks at 8 h during IgE sensitization. • Distinct temporal changes indicate different stages of membrane and mechanical remodeling during IgE sensitization. • Nanoscale characterization of extracellular vesicles shows distinct morphological features. • Adhesion and stiffness provide label-free mechanical indicators of cellular state in RBL-2H3 cells.
