• The review highlights the increasing importance of mannan conjugates in various domains, including vaccine delivery, therapeutics and drug delivery, diagnostics, and tissue engineering. • It describes various fabrication strategies in which these natural polysaccharides are combined with drugs, carriers, and biomolecules (such as proteins and antigens) via reductive amination, aminoxyligation, carbodiimide coupling, mannosylation, and cyanylation. • Mannan–antigen or mannan–drug conjugates are effectively absorbed, processed, and expressed on dendritic cells and macrophages. Thus, they have potential to be used successfully in the management of various diseases such as tuberculosis, Alzheimer’s disease, cancers, and fungal infections. • This review addresses the promise presented by the biocompatibility, biodegradability, and non-toxicity of this natural polysaccharide.
