• Tick-inspired barbed microneedles ensure robust skin anchoring and deep delivery. • HAMA-PBA matrix ensures ROS-responsive, on-demand release of TXA and ADSC-Exos. • ADSC-Exos uniquely serve as dynamic cross-linkers and potent ROS scavengers. • TXA and ADSC-Exos exert asymmetric synergy to remodel the dermal microenvironment. • This platform achieves unprecedented depigmentation in a severe melasma mouse model.
