• The secretome of Phanerochaete chrysosporium effectively biotransforms the fluoroquinolones levofloxacin and danofloxacin (5–25 mg/L). • LiPs were identified as the key enzymes driving degradation, achieving up to 100% removal under optimized conditions. • Mn²⁺ ions were found to partially inhibit LiP activity. • LC–MS/MS analysis revealed transformation into dealkylated, hydroxylated, and oxidized derivatives. • Toxicity assay indicated that the generated by-products exhibited significantly reduced toxicity compared to the parent antibiotics.
