Abstract

• We propose a new method for optical spatial differentiation and quantitative phase microscopy based on the birefringence effect of a 0.4 mm-thick calcite crystal. • We establish a differential interference measurement theory based on the birefringence effect, which serves as a robust method for high-precision measurement of minute displacements. • The system complexity was reduced and the imaging efficiency was improved by using thin plate crystals and unidirectional gradient phase recovery methods.
