• Higher urinary BPA exposure was associated with greater stroke prevalence. • Shared BPA-stroke targets converged on vascular and inflammatory pathways. • ESR1, NOS3, and F13A1 were prioritized as candidate BPA-stroke genes. • Single-cell analysis localized BPA-stroke signatures to activated endothelium. • BPA-treated HUVECs showed changes in endothelial dysfunction-related markers.
