• Assessed defensive enzymes (PAL, LOX, CHT) and secondary metabolites (phenol, MDA) in eleven rice genotypes under Pyricularia oryzae stress. • Gene expression analysis confirmed that BRRI dhan74 along with checkline IR 64 upregulated OsLOX2, OsCHT, and OsPAL, while BRRI dhan63 showed downregulation. • A total of 55 defense-related genes (30 CHT, 16 LOX, and 9 PAL) were identified, indicating a complex defense repertoire in rice. • Multi-omics approach integrates biochemical, molecular, and genomic characters to identify targets for blast resistance.
