• Basement membrane (BM) components are described in several resources, but information about their interactions is lacking. • Systems biology approaches address the challenge of harnessing data by applying techniques such as network analysis and computational modelling. • We describe key elements of BM structure, composition and dynamics together with the application of computational modelling. • Once BM organising principles have been defined, disease models can be developed showing how the loss of specific BM components leads to dysfunction.
