• Mn oxidation by bacterial multicopper oxidase Mnx in the presence of Ca2+ is studied. • Ca modestly affects the first step of the reaction, Mn(II)-Mn(III) oxidation. • Ca enhances the second step, disproportionation of Mnx-bound dimeric Mn(III). • The acceleration is cooperative in nature, revealing that two Ca2+ act together. • One Ca binds for every 2 Mn in the proposed Mnx's initial product, a MnO2 tetramer.
