• Cadmium pre-exposure significantly enhances JEV infection in both in vivo and ex vivo models. • Cadmium impairs innate antiviral signaling by disrupting the JAK/STAT axis. • Poly(I:C)-induced antiviral responses are markedly attenuated following cadmium pre-exposure. • Rather than acting on viral particles, cadmium primes host cells into an immunologically permissive state.
