Cascade Dual-Channel SPR Fiber Sensor Based on Composite Hydrogel Self-Assembled-Layers for High-Precision Detection of Glyceryl Tributyrate and Temperature

Glyceryl tributyrate is an ester compound that plays a crucial role in maintaining human intestinal health, immune regulation, and metabolic control. This study introduces a lipase-based specific self-assembled layer optical fiber SPR sensor, which achieves dual-parameter detection of glyceryl tributyrate and temperature for the first time. The assembly layer utilizes polyacrylic acid (PAA) and chitosan (CS) to immobilize lipase in the detection area of the sensor, achieving specific detection. MoS<sub xmlns:mml="http://www.w3.org/1998/Math/MathML" xmlns:xlink="http://www.w3.org/1999/xlink">2</sub> is employed to detect temperature, allowing for modulation and differentiation of the dual-channel operating wavelengths and enhancing the sensor’s sensitivity. Initially, simulations were conducted using COMSOL to clarify the sensor’s working principle. Subsequently, detection spectra under 0-6 layers were analyzed at detection concentrations ranging from 0.5 to 5 mmol, ultimately selecting 2 layers as the detection layer count. Then, single-channel temperature detection was performed on the temperature channel. Finally, the dual-channel sensor underwent cascade operation detection, along with stability and specificity tests. The results showed that at detection concentrations ranging from 0.5 to 5 mmol, the sensitivity of the concentration probe was 3.448 nm/mmol, with a detection limit of 0.32 mmol, meeting the millimeter-scale standards required for anticancer applications. The temperature probe exhibited a sensitivity of −2.43 nm/°C over the range of 0–100 °C. Under dual-channel cascade operation, both detection spectra could operate independently, enabling simultaneous detection of temperature and glyceryl tributyrate. This sensor offers advantages such as high sensitivity, simplicity, and label-free measurement, providing a novel approach for simultaneous detection of glyceryl tributyrate and temperature.
