Identifying key nodes within multi-layer GRNs is crucial for uncovering potential biomarkers and therapeutic targets. Key nodes exhibit both local structural advantages (e.g., bridging weak connections, disease associations) and global influence (e.g., centrality, regulatory impact). To comprehensively quantify node importance in multi-layer GRNs, we propose the CBFI (Comprehensive Biological Feature-based Importance) algorithm.
