• Conformational changes in TG2 can mask autoantibody epitopes. • Celiac disease plasma cells preferentially recognize the active enzyme conformation. • Substrate binding to TG2 does not affect plasma cell reactivity. • mAbs from naïve B cells are more conformation sensitive than mAbs from plasma cells.
