• Wharf roaches (Ligia spp.) ingesting EPS were studied by RNA-seq and metagenomics. • EPS feeding up-regulated host xenobiotic-metabolism enzymes in the gut. • Overall gut microbial diversity (alpha and beta) was unchanged by EPS exposure. • A few archaeal and viral taxa were detected only in the EPS group. • Wharf roach is proposed as a tractable model for gut-level plastic-exposure studies.
