• Develop a chemogenetic screening platform to identify synthetic lethal compounds targeting MYC. • Identify ALW-II-41-27, an EphA2 inhibitor, as a top hit with strong MYC-selective cytotoxicity. • ALW-II-41-27 induces MYC-dependent apoptosis via the intrinsic apoptotic pathway, independent of p53 status. • In vivo, ALW-II-41-27 inhibits tumor growth in MDA-MB-231 and MDA-MB-468 TNBC xenografts without apparent toxicity.
