• Development of chitosan-shielded exosomes (cExo) via electrostatic surface assembly. • Significant enhancement of colloidal and lyophilization stability for long-term storage. • Enhanced paracellular transport of exosomes via transient tight junction modulation. • Two-fold increase in dermal penetration confirmed by ex-vivo porcine skin assays. • Synergistic antioxidant and wound-healing effects for advanced skin-booster applications.
