• Laurdan and Cholesterol-pyrene reveal different behavior of membrane bilayer. • Lipid order and mobility are not simply correlated depending on molecular spaces. • The ordering effect of sterols is different in phosphatidylcholine and sphingomyelin. • In sphingomyelin, hydroxysterols show strong Lo character near the head-groups. • In sphingomyelin, hydroxysterols show low Lo character near the membrane center.
