• Combined prenatal stress and MIA produces comprehensive schizophrenia-like deficits in mice. • Dual-hit offspring show impaired PPI, social behavior, and increased immobility. • Clozapine treatment during the adolescent period fully reverses behavioral abnormalities. • Prefrontal Sptan1 downregulation and Glul upregulation identified in MODEL group. • Adolescent treatment window may prevent progression of neurodevelopmental pathology.
