• We developed a colorimetric assay based on micro-sized beads with ferrihydrite for on-site therapeutic drug monitoring. • The micro-sized beads incorporating ferrihydrite possess enzyme-like catalytic activity and an intrinsic green coloration. • The coating of microbeads with an amphiphilic polymer effectively prevented nonspecific adsorption of contaminating proteins. • Adsorption of the analyte onto the beads suppressed the color change, and the color decreased with the analyte concentration.
