• V. flaviceps and G. rubrolineatum exhibited the highest expression level of defensin. • Defensins exhibited distinct phylogeny from that of typical hymenopteran and heteropteran lineages. • Only PGRP-SC2 showed an evolutionary pattern consistent with hymenopteran lineage relationships. • Defensin of G. rubrolineatum possessed the strongest Gram-positive bacteria-specific anti-microbial activity.
