• Three rat caudal IDD models, PNP, BI and MFA, were directly compared at the same disc level. • X-ray, MRI, histology, IF and IHC were used to assess structural and matrix changes in each model. • BI induced the fastest and most severe degeneration, with disc height loss, MMP13 increase and COL2A depletion. • PNP caused moderate-to-severe degeneration, while MFA showed slower and milder matrix degeneration. • These data guide model selection for late-stage IDD, early degeneration and regenerative therapy studies.
