• Xanomeline and ketamine showed both shared and distinct molecular networks in MDD. • EGFR, IGF1R, and SRC were identified as common core targets of both drugs. • Xanomeline was linked to receptor tyrosine kinase/PI3K–AKT signaling, whereas ketamine was linked to glutamatergic/NMDA-related pathways. • Docking analysis supported partial convergence on shared plasticity-related signaling nodes
