• Clinically significant effects of either genetically predicted higher GLP-1 and/or lower GIP levels concerning cardio renal liver metabolic diseases were observed. • Joint genetic elevation of GLP-1 and reduction of GIP levels result in complementary and additive beneficial effects, conferring benefits in terms of body weight regulation and cardio renal liver metabolic diseases. • Low-level GIP may exert its cardiovascular benefits primarily through reducing body weight.
