• We established an in vitro infection model for DIV1 using mosquito cells, systematically determining the optimal virus-cell incubation time to be 6 h post-infection. • DIV1 propagated in the established cell model reached a titer of 10−2.67 TCID50 per 0.1 mL and induced characteristic cytopathic effects, including cell rounding, granulation, and detachment. • Cell-cultured DIV1 retained high pathogenicity in Macrobrachium rosenbergii, causing dose-dependent mortality and severe histopathological damage in the hepatopancreas, with visible viral particles detected in affected tissues. • Complete genome of a DIV1 strain was sequenced (165,567 bp, 34.58% GC, 176 ORFs), confirming its classification within the genus Decapodiridovirus.
