• A cost-effective glucose biosensor was fabricated on a 316L stainless steel wire substrate. • Graphene incorporation enhances electrochemical sensitivity but can reduce linear response. • PVDF-HFP polymer coating is shown to significantly improve analytical linearity (mean R2 > 0.92). • Sensor performance is governed by synergistic and antagonistic interactions among additives. • A systematic comparison of twelve distinct functional sensor architectures is presented.
