• Nanopatterned gold-coated quartz crystal sensors were elaborated by EBL. • A 23mer DNA aptamer was chemisorbed onto the gold surface. • QCM-D and SERS provided the sensing layer’s molecular composition and the mass uptake. • Binding of streptomycin was concomitantly monitored by QCM-D and SERS. • Dissociation constant was deduced from frequency shifts recorded upon streptomycin binding.
