Cross-priming underlies the efficacy of antibody–drug conjugates and immunotherapy combinations

Several recent preclinical and clinical studies support synergistic, or at least additive, effects between immunogenic cell death-inducing chemotherapy and anti-programmed death-ligand 1 checkpoint inhibitors. Antibody–drug conjugates are gaining ground in the clinic for solid and hematological malignancies, and recent data show antibody–drug conjugates are able to elicit tumor-cell demise with key features of immunogenic cell death. Antigens from tumor cells killed by antibody–drug conjugates are cross-presented by conventional type-1 dendritic cells, leading to cross-priming of cytotoxic T-lymphocyte responses. Immunogenic cell death enables cross-priming by conventional type-1 dendritic cells, effectively bridging innate and adaptive antitumor immunity. Conventional type-1 dendritic cells cross-priming functions point to mechanisms that can be synergistically exploited by novel antibody–drug conjugate-plus-immunotherapy combinations.
