• MeJA-responsive CsMYB6 is a key regulator in saffron crocin biosynthesis. • CsMYB6 overexpression boosts crocin content in Crocus sativus stigmas. • CsMYB6 interacts with crucial enzyme gene promoters (CsZ-ISO3 and CsCCD2) involved in crocins biosynthesis. • Proposed “JA-CsMYB6-CsZ-ISO3/CsCCD2-crocins” model sheds light on crocins biosynthesis regulation.
