• Brain-mimetic hydrogels reproduced nonlinear and viscoelastic mechanical features characteristic of brain tissue. • Melanoma cell behavior was dependent on plasticity states (MITFhigh/AXLhigh) and metastatic origin. • Decreasing matrix stiffness promoted proliferation and cluster formation in a cell state–dependent manner. • Hyaluronic acid emerged as a key regulator of melanoma cell spreading in brain-like microenvironments. • Brain-derived melanoma cells exhibited enhanced spreading and adaptation to biomimetic extracellular matrices.
